1. Yes, Destiny of Velious is a separate purchase. I would suggest holding off on getting DoV until you have a character that is at least level 85+ in either adventuring or tradeskilling.
2. Currently, the maximum amount of AAs is 320, if you have purchased Age of Discovery (300 if not). This amount normally increases with each expansion.

Well, isn't the AA cap still at 250 for people that don't have DoV or AoD?

Ah yes, forgot that DoV isn't standard. 250 with Sentinel's Fate, 300 with Destiny of Velious, 320 with Age of Discovery and Destiny of Velious.
